Chromatin immunoprecipitation followed by quantitative PCR was performed to validate the direct binding interaction between transcription factor TFEB and the promoter region of Thrombospondin-1 (Thbs1). This experiment was conducted to confirm computational predictions that TFEB directly regulates Thbs1 transcription. The assay demonstrated that TFEB physically binds to the Thbs1 promoter region, establishing a direct transcriptional regulatory relationship. This finding was crucial for understanding the mechanism by which TFEB modulates cardiac fibroblast behavior through Thbs1-mediated signaling pathways.
